John C. McGinley was born in Greenwich Village, New York in 1959. His love for acting began as an undergraduate at New York University. In 1984, he received a master of fine arts degree from the New York University Theater Program.
McGinley has appeared in the films Wall Street, Identity, The Animal, Any Given Sunday, Wagons East, The Rock, Nothing to Lose, Se7en, Office Space, Platoon, Are We Done Yet? and Wild Hogs. McGinley has also served as producer on a number of films including Colin Fitz and Watch It.
In his Los Angeles home, McGinley spends a lot of time with his young son, Max, who has Down Syndrome, and he is actively involved in fund-raising activities for the National Down Syndrome Society (NDSS). McGinley serves as the 2004-05 national spokesperson for the Buddy Walk, an advocacy walk organized by the NDSS.

- John enjoys surfing, weightlifting and softball.
- It was John's idea to have Dr. Cox call J.D girl's names, reflecting a habit he has with real life friends.
- Also, Dr. Cox's whistling to get peoples' attention and his nose scratches were all improv by John, but they caught on and became part of the character.
- He's often refered to as "Johnny C." by fellow cast members.
